Description:Brucellosis, a zoonotic disease caused by Brucella bacteria, remains a public health concern. The various biovars of Brucella abortus exhibit distinct virulence and host adaptation patterns. Metabolomics, a powerful tool for identifying metabolic signatures, holds promise for biovar differentiation. This study aimed to differentiate between six Brucella abortus biovars using UHPLC-Q-Orbitrap metabolomics in full scan and parallel reaction monitoring acquisition modes. Intracellular metabolites were extracted from each biovar employing acetonitrile:isopropanol:water. Unsupervised analysis (PCA, HCA and Volcano Plots) revealed distinct metabolite profiles among the biovars, with biovar 3 showing the most significant difference. The suggested ten biomarkers were correlated with amino acid synthesis, vitamin B6 metabolism, biofilm formation, and purine biosynthesis. This study is the first to explore metabolomics for Brucella abortus biovar differentiation. Further studies are needed to validate these findings and explore the potential biomarkers for diagnosis or drug development.
