Description:The integration of internal metabolism with external environmental cues is essential for the successful establishment and survival of seedlings. Light is an important mediator of the transition of skotomorphogenesis to photomorphogenesis. Understanding of this metabolic adaptation in barley, a major global cereal crop, is important for both fundamental plant biology and agricultural resilience. Currently, comprehensive metabolomic profiles of this time-dependent transition in the developing tissues of barley germlings/seedlings, are lacking. This study aimed to systematically map the temporal and tissue-resolved metabolic reprogramming that occurs in barley (Hordeum vulgare L.) seedlings grown under light/dark photoperiod and in complete darkness. Untargeted metabolomics based on UHPLC-MS was used to characterise the seeds, shoots and roots of barley germlings at 3-, 6- and 9-days post imbibition under a normal photoperiod (16h light/8h dark) or in complete darkness.
