Description:Radiation-induced liver damage (RILD) is a disease characterized by a series of physiological and pathological changes in liver tissue following exposure to a certain dose of radiation, and it is also a common complication of liver cancer and abdominal tumor radiotherapy. To date, the pathogenesis of RILD remains unclear, and effective diagnostic and therapeutic approaches are lacking. Based on this, the present study established an animal model of radiation-induced liver disease (RILD) using whole-liver irradiated rats. Metabolomics and transcriptomics were integrated to analyze liver tissue samples collected 7 days post-irradiation. The involved metabolic disorders primarily include ammonia metabolism, amino acid metabolism, glutathione metabolism and lipid metabolism. Moreover, a panel of potential plasma metabolic markers was identified through correlation analysis between liver tissue and plasma metabolic characteristics. Subsequently, the levels of radiation injury within 7 days post irradiation were assessed. This study provides experimental evidence for the identification of early diagnostic markers for whole-liver irradiation and RILD, as well as for exploring the molecular and pathophysiological mechanisms of RILD.
