Description:Previously uploaded CosMx Transcriptomics primary datasets SNT765.QQMH.597 and SNT733.NCTK.452 The brain experiences age-associated molecular, structural, and functional changes that coincide with greater incidence of neurological disease. Senescent and inflammatory cells may contribute to the maladaptive remodeling and reprogramming of aging brain parenchyma and are potential mediators of age-dependent brain dysfunction. To detect senescent cells and their gene signatures with preserved spatial context, we employed CosMx spatial molecular imaging (SMI) transcriptomics to analyze the distribution and biological heterogeneity of cell populations in aged mouse brain tissue slices. Sagittal 6 μm sections of corticolimbic regions from 24 month-old C57BL/6 female mouse brain were used to generate CosMx SMI data (120 nm / pixel resolution), corresponding to morphological staining with DAPI, histone, S6 RNA, and GFAP immunolabel markers and 1000-plex gene transcript localizations using the Mouse Neuro Panel. A custom 50-gene senescence-associated probe set was incorporated to detect senescence signatures across the brain. Morphology markers enabled accurate cellular segmentation, while transcript coordinates were assigned three-dimensional spatial positions within polygonal cell boundaries. Each field-of-view (FOV) image measured 500 x 500 μm, and individual FOVs were tiled to generate contiguous, large-area sections per sample.
