Description:We aim to map the distribution and transcriptional heterogeneity of senescent cells in skeletal muscle tissue of aged mice using Xenium spatial transcriptomics. We use p21-Cre mice to eliminate p21high senescent cells in vivo. The skeletal muscle tissue is collected from three groups of mice: young mice (6 months old), aged mice (27 months old, p21-Cre/+; LUC/+), and aged mice with eliminated senescent cells (27 months old, p21-Cre/+; DTA/+). After fixation, paraffin embedding, and sectioning, tissues are analyzed using Xenium spatial gene expression profiling and stained with hematoxylin and eosin (H&E). Spatial mapping visualizes tissue structure and the localization of senescent cells, comprehensively demonstrating how the number, characteristics, and microenvironment of senescent cells change in skeletal muscle tissue.
