Description:The overall objective of SenNet-PNNL TDA project is to significantly advance our microfluidics-based spatial proteomics platform, termed laser capture microdissection coupled with nanodroplet processing in one-pot for trace samples (LCM-nanoPOTS), and apply this technology to map SNCs and their SASP in different mouse and human tissues. In the UG3 phase, we have established a high-throughput and robust single-cell isolation system and coupled it with nanoPOTS-MS. We have modified the Zeiss LCM system to enable reliable single-cell isolation and collection by designing and assembling a robot-addressable capillary sampling probe. To demonstrate the capability of this spatial single-cell proteomics platform, we used the stromal senescence mouse model (FASST mouse, FibroblastsAccelerate Stromal-Supported Tumorigenesis) from Dr. Prof. Megan Ruhland group at OHSU. It is induced a large number of senescence cells (SNCs) in 4-week-old FASST mice with tamoxifen treatment and FASST mice show increased stromal senescence within the skin. FASST mouse and control skin tissue sections were cut at 10 μm thickness onto PEN membrane-coated slides. The tissue was stained for senescence-associated beta-galactosidase activity to identify SNCs.For mapping of SNCs and their SASP in mouse skin tissue. This data set contains our first spatial single cell proteomics set from the FASST mouse. : ./1244831_NanoPOTS_LA_MouseSkin_cre_Pos_nonsenescence_5CellS_C6
