Description:Cellular senescence and its role in aging and age-related disease is still unclear, despite the recent advances in recognizing aging as a risk factor for many chronic illnesses. In the kidney, aging is associated with molecular and physiological changes that cause a loss of renal function and of regenerative potential. In mice, kidney aging is accompanied by characteristic changes in the glomerulus leading to overall decline in glomerular structure and function with advancing age. To build a comprehensive and comparative map of senescence in the mice kidney we applied Visium spatial transcriptomics in young and aged mice of both sexes (6 and 24 months C57BL/6J; n=3 per group). With a spatial approach we were able to analyze healthy glomerulus, hyalinized glomerulus, and accumulation of inflammation directly from tissue section of mouse kidney samples. We also generated digitized high-resolution images of hematoxylin and eosin (H&E)-stained kidney cross sections for comparation. We have implemented computational analysis approaches to characterize age- and senescence-related transcriptomic changes in the mouse kidney.
